Chloramphenicolacetyltransferase(CAT),encodedbyabacterialdrug-resistancegene,inactivateschloramphenicolbyacetylatingthedrugatoneorbothofitstwohydroxylgroups.Thisgeneisnotfoundineukaryotes,andthereforeeukaryoticcellscontainnobackgroundofCATactivity.Thischaracteristic,alongwiththeeaseandsensitivityoftheassayforCATactivity,hasmadetheCATgeneoneofthefirstreportergenesusedforstudiesofmammaliangeneexpression.

LinkageofputativeregulatorysequencestotheappropriatepCAT^®3ReporterVectorandsubsequenttransfectionallowsfortheefficientassayofCATactivityinculturedcells.

CATactivitymaybemonitoredbytwoalternativemethodsintheCATEnzymeAssaySystem.Themostrapid,sensitiveandconvenientoftheseassaysisbasedonliquidscintillationcounting(LSC)ofCATreactionproducts.CATactivityalsocanbeanalyzedusingthin-layerchromatography(TLC).Thismethodismoretime-consumingthantheLSCassaybutallowsvisualconfirmationofthedata.
