1.Placeoneneedleofsolidbacterialgrowthortwoloopsofliquidbacterialgrowthinthecenterofacleanslide.2.Ifworkingfromasolidmedium,addonedrop(andonlyonedrop)ofwatertoyourspecimenwithawaterbottle.Ifusingabrothmedium,donotaddthewater.3.Now,withyourinoculatingloop,mixthespecimenwiththewatercompletelyandspreadthemixtureouttocoverabouthalfofthetotalslidearea.4.Placetheslideonaslidewarmerandwaitforittodry.Thesmearisnowreadyforthestainingprocedure.
