VEGFwasinitiallypurifiedfrommediaconditionedbynormalbovinepituitaryfolliculo-stellatecellsandbyavarietyoftransformedcelllinesasamitogenspecificforvascularendothelialcells.ItwassubsequentlyfoundtobeidenticaltoanindependentlydiscoveredvascularpermeABIlityfactor(VPF),whichwaspreviouslyidentifiedinmediaconditionedbytumorcelllinesbasedonitsabilitytoincreasethepermeabilityofcapillarybloodvessels.ThreemouseCDNAclones,whicharisethroughalternativesplicingandwhichencodematuremousemonomericVEGFhaving120,164,or188,aminoacids,respectively,havebeenidentified.Tworeceptortyrosinekinases(RTKs),Flt-1andFlk-1(themousehomologueofhumanKDR),bothmembersofthetypeIIIsubclassofRTKscontainingsevenimmunoglobulin-likerepeatsintheirextracellulardomains,havebeenshowntobindVEGFwithhighaffinity.TherolesofthehomodimersofKDR,Flt,andtheheterodimerofKDR/FltinVEGFsignaltransductionremaintobeelucidated.Invivo,VEGFhasbeenfoundtobeapotentangiogenesisinducer.
