Glutathioneisatripeptide(g-glutamylcysteinylglycine)andexistsinreduced(GSH)andoxidized(GSSG)states.GlutathioneperoxidasecatalyzestheformationofGSSGfromGSH,andglutathionereductaserecyclesGSSGtoGSH.Withincreasedlevelsofoxidativestress,intracellularGSSGwillaccumulate,andtheGSH/GSSGratiowilldecrease.Therefore,thedeterminationoftheGSH/GSSGratioisausefulindicatorofoxidativestressincellsandtissues.Thiscolorimetricassayisbasedonthereactionbetween5’,5’-Dithiobis2-nitrobenzoicacid(DTNB)andGSHtoformTNB,whichexhibitsmaximumabsorbanceat412nm.TheintensityoftheabsorbanceisproportionaltotheGSHlevelinthesample.
