GM‑CSFwasinitiallycharacterizedasafactorthatcansupporttheinvitrocolonyformationofgranulocyte‑macrophageProgenitors.Itisalsoagrowthfactorforerythroid,megakaryocyte,andeosinophilprogenitors.GM‑CSFisproducedbyanumberofdifferentcelltypes(includingTcells,Bcells,macrophages,mastcells,endothelialcells,fibroblasts,andADIpocytes)inresponsetocytokineorinflammatorystimuli.Onmaturehematopoieticcells,GM‑CSFisasurvivalfactorforandactivatestheeffectorfunctionsofgranulocytes,monocytes/macrophages,andeosinophils(1,2).GM‑CSFpromotesaTh1biasedimmuneresponse,angiogenesis,allergicinflammation,andthedevelopmentofautoimmunity(3‑5).Itshowsclinicaleffectivenessinamelioratingchemotherapy‑inducedneutropenia,andGM‑CSFtransfectedtumorcellsareutilizedascancervaccines(6,7).The22kDaglycosylatedGM‑CSF,similartoIL‑3andIL‑5,isacytokinewithacoreoffourbundledalpha‑helices(8‑12).MaturehumanGM‑CSFshares63%‑70%aminoacidsequenceidentitywithcanine,feline,porcine,andratGM‑CSFand54%withmouseGM‑CSF.GM‑CSFexertsitsBIOLOGicaleffectsthroughaheterodimericreceptorcomplexcomposedofGM‑CSFRalpha/CD116andthesignaltransducingcommonbetachain(CD131)whichisalsoacomponentofthehigh‑affinityreceptorsforIL‑3andIL‑5(13,14).Inaddition,GM‑CSFbindsanaturallyoccurringsolubleformofGM‑CSFRalpha(15).HumanGM‑CSFisactiveoncanineandfelinecellsbutnotonmurinecells(16‑18).

Western Blot: NOD2 Antibody (2D9) [NB100-524] - Total protein from human THP1 and US-OS cells and mouse Raw264.7 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-NOD2 in 1% non-fat milk in TBST and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
Immunocytochemistry/Immunofluorescence: NOD2 Antibody (2D9) [NB100-524] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-NOD2 (2D9) NB100-524 at a 1:200 dilution overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was counterstained with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Strong bands at 40, 45kDa correspond to DCX protein, detected only in the developing brain.
Immunocytochemistry/Immunofluorescence: Doublecortin Antibody (3E1) [NBP1-92684] - Analysis of cortical neuron-glial cell culture from E20 rat stained with mouse mAb to Doublecortin (DCX), NBP1-92684, dilution 1:1,000 in red, and costained with chicken pAb to Microtubule Associated Protein2 (MAP2), dilution 1:10,000 in green. The blue is DAPI staining of nuclear DNA. DCX antibody reveals strong cytoplasmic staining in a population of small developing neurons and their processes, while MAP2 antibody stains dendrites and perikarya of mature neurons. Therefore DCX antibody is an excellent marker of developing neuronal cells.
Immunocytochemistry/Immunofluorescence: Doublecortin Antibody (3E1) [NBP1-92684] - Rat brain neural cultures stained with NBP1-92684 (green), a chicken polyclonal antibody to GFAP (NB300-213, red) and DNA (blue). The NBP1-92684 antibody reveals strong cytoplasmic staining in a population of small developing neurons. These cells are often found in small clumps as in these cultures, as in this example. Note that they are not positive for MAP2, which is characteristic of more mature neurons, and that the mature neuron shown is negative for NBP1-92684. This Doublecortin antibody is therefore an excellent marker of developing neuronal cells.
