·Holdingthesyringecomfortably,withoneconcertmotioninserttheneedleintotheheartthroughthediaphragmanddrawblood

·Shouldthefirstattemptnotsucceedandpneumothoraxdevelops,immediatelyopenthechestcavitythroughamidlineincision(sternotomy)anddrawblooddirectlyfromtheheart

·Spinat1000rpm(450g)for10minutesat4°C

·Aspiratesupernatanttopalletusingvacuumsetup

·Add2mloflyticbuffersolutionforthesecondtimefortheresidualRBCsandfilltubeswithPBS.Spinagainfor10minutesat1000rpmat4°C

·Aspiratesupernatantwithvacuumsetuptopallet

·Add?mlofPBStothepalletsandgentlymixthem

·Specimensarereadytobereadbythefacsmachine.Theycanbestoredat4°Cforatleast2days.
