TheAssaySenseHumantPAChromogenicActivityKitisdevelopedtodeterminehumantPAactivityinplasmaandcellculturesupernatants.TheassaymeasurestheabilityoftPAtoactivatetheplasminogentoplasminincoupledorindirectassaysthatcontaintPA,plasminogen,andaplasmin-specificsyntheticsubstrate.Theamountofplasminproducedisquantitatedusingahighlyspecificplasminsubstratereleasingayellowpara-nitroaniline(pNA)chromophore.ThechangeinabsorbanceofthepNAinthereactionsolutionat405nmisdirectlyproportionaltothetPAenzymaticactivity.
