MouseCCL2isamemberoftheβ(CC)subfamilyofchemokines.ThemouseCCL2genewasinitiallyidentifiedasaplateletderivedgrowthfactorinduciblegeneinmousefibroblasts.MouseCCL2cDNAencodesa148aminoacid(aa)residuewithaputative23aasignalpeptidethatiscleavedtogeneratethematureprotein.MouseCCL2shares82%aminoacidsequenceidentitywithratCCL2.MouseCCL2alsoshares55%aminoacidsequenceidentitywithhumanMCP1.ComparedtohumanMCP1,mouseCCL2hasa49aaresidueextensionatthecarboxyterminus.WhenaDNAsequenceencodingthe125aaresidueofthematureCCL2proteinwasexpressedinE.coliatR&DSystems,thepurifiedproteinhadthepredictedN-terminusbutamassof8525Da.ThetruncationofmostoftheCterminalextensioncouldbedueeithertopurificationartifactortoposttranslationalmodification.ThetruncatedrecombinantCCL2hasapotencysimilartothatofhumanMCP1inthemonocytechemotaxisassay.MouseCCL2hasfullactivityonhumancellswhilehumanMCP1haslimitedactivityonmousecells.
