   TESfreeacidisverysolubleinwater;T1375,routinelytestedforsolutionappearanceat33%(w/v),givesaclearcolorlesssolution.T6541,SigmaUltra,givesaclearcolorlesssolutionat1M,withapHrangeof3.5-5.0.3Solutionsshouldbestableat2-8ECforatleastsixmonths.Sterilizationshouldbedonebyfiltrationthrough0.2mfilters,sincegenerallyautoclavingisnotrecommendedforanysulfonicacidbuffers.However,autoclavabilitywascheckedon0.5MsolutionsatpH6.7,7.45and8.5,andat0.05MatpH7.4.ThepHwasthesamebetweenautoclavedandnon-autoclavedsamplesandsolutionsremainedcolorless.

GENERALREMARKS:
   TESisastructuralanalogtoTrizmabuffer,oneoftheethanesulfonicacidseriesofbiologicalbuffersdevelopedbyGoodetal.,tomeetthesecriteria:midrangepKa,maximumwatersolubilityandminimumsolubilityinallothersolvents,minimalsalteffects,minimalchangeinpKawithtemperature,chemicallyandenzymaticallystable,minimalabsorptioninvisibleorUVspectralrange,andeasilysynthesized.Althoughthestructureaboveisdrawnasaneutralmolecule,itprobablyexistsinsolutionasazwitterion.
   BecausethepKais7.4(physiologicalpH)thisbufferhasconsiderablepotentialforavarietyofbiologicalapplications.Inmanyculturemediathatrequiremetalcations,manybufferscannotbeusedduetochelationorprecipitation(citrateorphosphate,forexample.)TESwasfoundtobeparticularlybeneficialinastudyofsuccinateoxidation.
   AbufferusingTESfreeacidcanbepreparedbytitratingthefreeacid(T1375orSigmaUltraT6541)withsodiumhydroxidetothedesiredpH,usingaboutahalf-equivalentofNaOH.Alternatively,solutionsofequimolarTESfreeacidandTESsodium(T0772)canbemixedtoattainabufferofthedesiredpH.TitratingasolutionofTESsodiumwithHClisnotrecommendedsincetheresultingsolutionwillcontainahalf-equivalentofNaCl.
