Gendepot的T4 DNA连接酶是最通用的DNA克隆连接酶。这种ATP依赖性酶将5'-磷酸与3'-羟基化的末端共价连接，这些末端是通过限制性酶消化或其他酶促过程产生的双链DNA片段的平末端或相容的内聚末端。T4 DNA连接酶对单链核酸无活性。连接反应后，可通过在65℃温育10分钟使T4 DNA连接酶失活。
