Brevetoxinsarelipid-solublepolyetherneurotoxinsinitiallyproducedbythedinoflagellateKareniabrevis.BrevetoxinsactasagoNISTs,bindingtheneurotoxinsite5onSCN4A/Nav1.4andSCN5A/Nav1.5voltage-gatedNa+channelsinskeletalmuscleandcardiactissue,producingachannelopeningeffect;thisactivityresultsinbronchoconstrictionandairwayinflammationinvivo.Stimulationofthevoltage-gatedNa+channelsalsoinducesNMDAreceptor-mediatedreleaseofL-glutamateandL-aspartateinneurons.ActivityatthesechannelsalsoactivatesPyk2andSrc,inducingdownstreamsignalingcascades.Brevetoxin2formsDNAadductswithcytidineinisolatedlungcellsandwithadenosineandguanosineinvivo,indicatingcarcinogenicactivity.
