UsingDMEM
DMEMisuniquefromothermediaasitcontains4timestheconcentrationofaminoacidsandvitaminsthantheoriginalEagle"sMinimalEssentialMedium.DMEMwasoriginallyformulatedwithlowglucose(1g/L)andsodiumpyruvate,butisoftenusedwithhigherglucoselevels,withorwithoutsodiumpyruvate.DMEMcontainsnoproteins,lipids,orgrowthfactors.Therefore,DMEMrequiressupplementation,commonlywith10%FetalBovineSerum(FBS).DMEMusesasodiumbicarbonatebuffersystem(3.7g/L),andthereforerequiresa5–10%CO2environmenttomaintainphysiologicalpH.
