ThepolyclonalantibodyrecognizesmouseC4d.ComplementfactorC4(MW41kDa),formerlyknownasGgprotein,consistsofanalpha-,beta-andgamma-chain.Theclassicalpathwayofcomplementandthemannosebindinglectin(MBL)activationpathwayconvergeatC4.ActivatedC1,MASP-1andMASP-2cleaveC4resultingintheformationofC4aandC4b.ThelattercanbecleavedbyfactorIresultinginC4candC4d,inwhichstepallfunctionalsitesarelost.TheC4dactivationfragmentofC4isanexcellentMarkerforclassicalcomplementpathwayandMBLpathwayactivation.ThethioesterformedbetweenthesidechainsofCys1010andGln1013withintheC4dregionoftheα-chainmediatescovalentattachmenttothetargetsurfacebearingactivatedformsofC1sorMASP.FurThermore,C4dishighlyhomologoustoC3dwithover35%sharedaminoacidsequence.Inanumberofdiseasessuchasrheumatoidarthritis(RA),hereditaryangioedema(HAE),systemiclupuserythematosus(SLE)andchronicurticariawithhypercomplementemialevelsofC4daresignificantlyelevatedinserumorplasma.C4dlevelsmayalsobeelevatedinplasmafrompatientswithavarietyofhumoralautoimmunediseasesinwhichcomplementactivationisknowntooccur.DepositionofC4dinperitubularcapillarieshasbeenshowntobeasensitivemarkerforantibody-mediated(humoral)rejectioninrenaltransplantbiopsies.
