ThemonoclonalantibodyM2139reactswiththeJ1epitope(triplehelicalposition551-564)ofcollagentypeII.Collagenisastructuralproteininbone,cartillageandconnectivetissue.CollagentypeII(CII)isthemajorcollagenofthenucleuspulposa(acomponentofspine),cartilageandvitreous(acomponentoftheeye).Themostcommonlyusedanimalmodelforrheumatoidarthritis(RA)isthecollagen-inducedarthritis(CIA).TransferofcollagentypeIIspecificmonoclonalantibodiesinducesanacuteformofarthritis(collagentypeIIantibodyinducedarthritis,CAIA).ThemonocolonalantibodyM2139hasbeenshowntoinduceCAIAinnaïvemiceafterinjectionoflipopolysacharide(LPS).However,incombinationwiththemonoclonalantibodyCIIC1,bindingtotheC1epitopeofCII,thepairofmonoclonalantibodiesinducearthritisindifferentstrainsofmicewithoutanyotherstimulants.Thepresenceofsecondarystimulus,LPS,increasesthediseaseincidenceandseverity.ThemonoclonalantibodyM2139iscrossreactivewithrat,bovine,chickenandhumanCII.
