ThemonoclonalantibodyCIICIreactswiththeC1epitope(triplehelicalposition358-364)ofcollagentypeII.Collagenisastructuralproteininbone,cartillageandconnectivetissue.CollagentypeII(CII)isthemajorcollagenofthenucleuspulposa(acomponentofspine),cartilageandvitreous(acomponentoftheeye).Themostcommonlyusedanimalmodelforrheumatoidarthritis(RA)isthecollagen-inducedarthritis(CIA).TransferofcollagentypeIIspecificmonoclonalantibodiesinducesanacuteformofarthritis(collagentypeIIantibodyinducedarthritis,CAIA).ThemonocolonalantibodyCIIC1hasbeenshowntoinduceCAIAinnaïvemiceafterinjectionoflipopolysacharide(LPS).However,incombinationwiththemonoclonalantibodyM2139,bindingtotheJ1epitopeofCII,thepairofmonoclonalantibodiesinducearthritisindifferentstrainsofmicewithoutanyotherstimulants.Thepresenceofsecondarystimulus,LPS,increasesthediseaseincidenceandseverity.ThemonoclonalantibodyCIIC1iscrossreactivewithrat,bovine,chickenandhumanCII.
