MASP-2isaMBLassociatedserineprotease.Mannan-bindinglectin(MBL)andficolins,incomplexwithMASPs,arecapableofactivatingthecomplementsystem,thusmediatingthedestructionofinfectiousagents.AfteroligomerisationofMBL(orL-Ficolin)MASP-2willbeboundandactivated.ActivatedMASP-2cleavesC4andC2andiscrucialfortheactivationofdownstreamcomplementcomponents.MASP-2deficiencywillresultinanincreasedsusceptibilityforinfections(e.g.leukemiapatientsinchemotherapyareathighriskofseriousinfections).EvenmoresothenMBLdeficiency,becauseMASP-2isalsorequiredforcomplementactivationbyH-andL-ficolin.TheELISAdetectsMASP-2inserumandplasma.TheMASP-2levelsinplasmafromhealthyindividualsrangefrom170to1196ng/ml.
