Thereactivityoftheantiserumisrestrictedtofibrinogen.InimmunoelectrophoresisandrADIalimmunodiffusion(Ouchterlony),usingvariousantiserumconcentrationsagainstnormaldogplasmaasingleprecipitinlineisobtainedwhichshowsareactionofidentitywiththeprecipitinlineobtainedwithpurifiedfibrinogen.Noreactionisobtainedwithanyotherplasmaproteincomponentorserum.However,theantiserummayalsoreactwithfibrinmonomers,circulatingfibrinopeptidesandfibrindegradationproducts.Inenzyme-immunocytochemicalandimmunohistochemicaltechniquesforthedetectionoffibrinogenatthecellularandsubcellularlevelinappropriatelytreatedcellandtissuesubstrates;asdetectionreagentinnon-isotopicmethodologyandsolidphaseimmunochemistry(e.g.ELISA,Westernblotting).Thisimmunoconjugateisnotpre-diluted.Theoptimumworkingdilutionofeachconjugateshouldbeestablishedbytitrationbeforebeingused.Excesslabelledantibodymustbeavoidedbecauseitmaycausehighunspecificbackgroundstainingandinterferewiththespecificsignal.

Fibrinogen(clottingfactorI)isaheatlABIlebetaglycoproteinpresentinplasma.Itistheprecursoroffibrin,whichisthekeyproteinconstitutingthenetworkofthebloodclot.Thrombinconvertsfibrinogentofibrinbylimitedproteolysis.Fibrinmonomerspolymerizetofibrinwhichisstabilizedbycross-linking.Fibrinogenisisolatedfromfreshplasmaafterremovingprothrombin.Freund’scompleteadjuvantisusedinthefirststepoftheimmunizationprocedure.
