BACKGROUND

FreerADIcalsarehighlyreactiveandgenerallyshort-livedspecies.Hence,moststudiesoffreeradicalshavebeenrestrictedtoanalysisofendproductsformedfollowingtheirinteractionwithcellularbiomolecules.Onetechnologythathaspermitteddirectstudyoffreeradicalsiselectronspinresonance(ESR).Forveryshortlivedradicals,spintrapshavebeenemployedtogeneratelongerlivedradicalsthataremoreamenabletoESRanalysis.Themostwidelyusedofthesespintrapsis5,5-dimethyl-1-pyrroline-N-oxide(DMPO).ThereactionofDMPOwithaproteinradicalgivesrisetoanitroxideradicalwhichcanbedisproportionatetohydrozylamineandnitroneadducts.Ofthesespecies,onlythenitroneadductcanbeconsideredstable.
