BACKGROUND

TransmissibleGastroenteritis(TGEV)isanenvelopedCoronaviruswithapositive-sensesingle-strandedRNAgenomeandahelicalsymmetry.Thegenomicsizeofcoronavirusesisapproximately28.6kilobases.ProteinsthatcontributetotheoverallstructureofTGEVincludethespike(S),envelope(E),membrane(M)andnucleocapsid(N).OthercoronavirusesbelongingtothisfamilyincludeFelinecoronavirus(FeCV),Caninecoronavirus(CCV),andFelineinfectiousperitonitisvirus(FIPV).TGEVinfectspigs.However,inyoungpigs,themortalityrateiscloseto100%.ThepathologyofTGEVissimilartothatofothercoronaviruses.Coronavirusesenterthehostbyfirstattachingtothehostcellusingthespikeglycoprotein.TheSproteininteractswiththeporcineaminopeptidaseN(pAPN),acellularreceptor,toaideinitsentry.AdomainintheSspikeproteinisrecognizedbypAPN,andtransfectionofpAPNoccurstononpermissivecellsandinfectsthemwithTGEV.Oncethevirusinfectsthehost,itmultipliesinthecellliningofthesmallintestineresultinginthelossofabsorptivecellsthatinturnleadstoshorteningofvilli.TheinfectedswinethenhasreducedcapABIlityfordigestingfoodanddiefromdehydration.
