ThereversIBLeconjugationofubiquitinandubiquitin-likeproteinstosubstratesregulatesalargenumberofcellularprocesses.Decipheringthesecomplexpathwaysisthefocusofmanyresearchersbuttheworkhasbeenslowedbythelackofhighqualityreagents.Deubiquitinatingenzymes(DUBs)recognizeandcleavetheisopeptidebondbetweenubiquitinmoietiesorbetweenubiquitin andthetargetprotein.MeasurementoftheactivityoftheseenzymesforkineticstudyorinhibitorscreeninghasbeentrADItionallycarriedoutusingubiquitinconjugatedtofluorescentleavinggroups(AMCorRhodamine)attheC-terminus.Whilethesearesuitablesubstratesformanydeubiquitinatingenzymes,thecleavageoftheamidebondservesasaproxyforthetrueactivityofisopeptidases,i.e.cleavageofanisopeptidebond.
LifeSensorshasproducedacompletepanelofalleightpossibledi-ubiquitinmolecules(includingCtoN-linkedlinearmolecules)fordeterminingthelinkagespecificactivityofindividualDUBs. Thiskitisaconvenientcollectionofthesedi-Ubmoleculesfortheaninitialsurveyoflinkage-specificity.
