IQFDiUbTechnologyLifeSensors’diubiquitinsubstratesrepresentanewclassofsubstratesforthecontinuousfluorescentmeasurementoftrueisopeptidaseactivity.TheC-terminusofwild-typeubiquitinisconjugatedviaanisopeptidebondtolysine11,48,or63ofasecondubiquitinmoleculewiththeresultantdiubiquitinforminganinternallyquenchedfluorescentFRETpair(IQF).Eachubiquitinislabeledwithasinglemoleculeofeitherafluorescentreporter(i.e.TAMRA)orahighlyefficientquenchingdye.CleavageoftheIQFDiUbbydeubiquitylasesleadstoseparationofthefluorophorefromquencherandsubsequentincreaseinobservedfluorescence.Theubiquitin-TAMRAreferencestandardcanbeusedtooptimizeandcalibratetheperformanceofindividualplatereaders(orfluorometers)formeasurementoftheratesofhydrolysisofourIQF-DiUbbyDUBs.
