Description:

Proteasomesareverylarge(20S,26S)proteinassembliesfoundinboththenucleusandcytoplasmofalleucaryotes(andinsomeprocaryotes).TheyareresponsIBLeforthedegradationandrecyclingofproteinswhichhavebeenpreviouslytaggedwithubiquitin.Suchtaggedproteinsaredegradedintopeptidesapproximately7-8aminoacidslongwhicharesubsequentlyfurtherdegraded.The20Sassemblyisthefunctionalproteasestructurewithchymotrypsin-like,trypsin-likeandcaspase-likeproteaseactivities.ProteasomeActivityAssaytakesadvantageofthechymotrypsin-likeactivity,utilizinganAMC-taggedpeptidesubstratewhichreleasesfree,highlyfluorescentAMCinthepresenceofproteolyticactivity.Thekitalsoincludesapositivecontrol(JurkatCelllysatewithsignificantproteasomeactivity)andaspecificproteasomeinhibitorMG-132whichsuppressesallproteolyticactivityduetoproteasomes.Thispermitsdifferentiationofproteasomeactivityfromotherproteaseactivitywhichmaybepresentinsamples.
