TRPM2channelbelongstothetransientreceptorpotentialchannel(TRP)superfamilywhichisdividedintosevenmainsubfamilies.TRPM2channelisanonselective,calcium-permeablecationchannelhighlyexpressedinthebrain,vascularsmoothmuscle,endothelialcellsandimmunecells.Itisactivatedbyoxidativestressorssuchashydrogenperoxide,cyclicADPribose,andNADP,leADIngtoanincreaseintheintracellularfreecalciumconcentrationandcelldeath.TRPM2isapotentialdrugtargetinavarietyofhumandiseases,includingcardiovascularandneurodegenerativediseases.

BIOLOGicalActivity:
N-terminalFLAGtaggedhumanTRPM2channelhasbeenstablyintegratedintoHEK293cellsanditsexpressioncanbeinducedbytetracycline(doxycycline).Thetetracycline-inducibleexpressionofTRPM2wasconfirmedbyWesternblotting.ThefunctionofTRPM2wascharacterizedbycalciumassay.TRPM2-HEK293cellswerepre-treatedwithdoxycyclinetoinducetheexpressionofTRPM2,thenstimulatedwithH2O2,andcalciuminfluxwasobserved.TheH2O2-inducedcalciuminfluxviaTRPM2couldbeblockedbyN-(p-Amylcinnamoyl)anthranilicAcid(ACA),aTRPchannelblocker.
