TypeICollagenisthemostabundanttypeofcollageninthehumanbodyandisamajorstructuralproteinprevalentinskin,bone,tendon,andotherfibrousconnectivetissues.Allcollagensarecharacterizedbyasecondaryproteinstructureconsistingofthreepolypeptidechainswrappedaroundoneanotherinatriplehelicalarrangement.Thedifferenttypesofcollagenvaryintheprimaryaminoacidsequenceoftheirpolypeptidechains,withtypeIcollagenbeingcomprisedoftwoidenticalα1(I)chainsandoneα2(I)chainthatspontaneouslyformthetriplehelixinsolution.CellAdhere™TypeICollagen,Human,Lyophilizedispreparedfromextracellularmatrixsecretedbynormalhumanfibroblastsandisxeno-free.Whenreconstitutedwith5mLof0.01NHCl,theresultingconcentrationis3mg/mL.ThisproductispurifiedusingahighlycontrolledmanufacturingprocesswithvalidatedstepstoensureinactivationofpossIBLeviralcontaminants,anditissterilitytested.Theendotoxinlevelis5.0EU/mLorless,asassayedbyLimulusamoebocytelysate(LAL)assay.Thisproductisidealforcoatingsurfaceswithathinlayerofproteintosupportcellattachment,butisnotsuitablefor3Dcellcultureapplications.Theoptimalproteinconcentrationmayvarydependingonthecelltypebeingused,andthereforemustbetitratedforbestresults.
