Hybridization Solution is a ready to use solution for nucleic acid hybridization. The solution is a proprietary formulation. Each lot of Hybridization Solution is tested by Northern and Southern hybridization to ensure quality.||Procedure:|1. Heat the solution at 65°C to dissolve the precipitate completely if there are precipitate formed at room temperature.|2. Prehybidize the membrane (Northern blot or Southern blot or RNA arrays) with solution at 65°C for 2-4 hours at the about amount of 0.2ml per cm2 membrane|3. After prehybridization, change the solution with fresh Hybridization Solution at 0.1ml per cm2 membrane, and keep at 65°C.|4. Heat your probe (either radioactive or non-radioactive probes is ok) at 95°C for 3-5 minutes, and transfer on ice immediately for 5 minutes.|5. Briefly spin down the probe, and add into the Hybridization Solution with the membrane. |6. Hybridize the membrane with the probe for from 2 hours to over night. |7. After hybridization, remove the solution.||For Radioactive Probe:|• Wash the membrane with 10 ml 2xSSC (0.05% SDS) for 10-15 minutes at room temperature for 3 times.|• Wash the membrane with 100 ml 0.1xSSC (0.1% SDS) for 20 minutes at 50°C for 2 times|• Expose the membrane to X-ray film for 24- 48 hours. ||For Non-radioactive Probe:|• Please follow the manufacturer’s instructions.
