DescriptionThetranscriptionfactorAP-1(activatorprotein-1)isinvolvedincellularproliferation,transformationanddeath.AP-1andnuclearfactorB(NFkB)canbespecificallytargetedtopreventcancerinductioninmousemodels.AP-1canbeproducedby18differentdimericcombinationsofproteinsfromtheJun[c-Jun(cat#P2014),JunBandJunD]andFos(c-Fos,FosB,Fra-1andFra-2)families,includingJunhomodimersandJun–Fosheterodimers(1–4).TheJunandFosproteinscontainabasic-regionleucinezipper(bZIP)domain,andarecapableofbindingtootherbZIPproteinsincludingthosefromtheATF,MAF,CNCandC/EBP(CCAAT/enhancer-bindingprotein)subfamilies(5).Jun–JunandJun–Fosdimersbindwithhighestaffinitytothe12-O-tetradecanoylphorbol-13-acetate(TPA)responseelement(TRE)[TGA(C/G)TCA](6),althoughmanyotherAP-1-likesiteshavebeenreported.Bindingtoanyofthesesitescanbetissue-specific,oraffectedbyneighboringsequences,anddependentuponinteractionswithothertranscriptionfactorsorcofactors.JunandFosproteinscanalsodimerizewithotherbZIPproteins,allowingthemtotargetotherDNAbindingsites,suchasthecAMPresponseelement(CRE),theantioxidantresponseelements(ARE),andhalf-sitescomposedofhalfofaTREsiteandhalfofaMAF-orCNC-bindingsite.Inaddition,AP-1proteinscaninteractwithotherproteins,includingthep65subunitofNF-B,CBP(CRE-binding-protein-bindingprotein)(p300),SMAD-3and-4,andtheretinoblastomaprotein(see5foramorecompletelist),furtherincreasingthecombinatorialpotentialofJunandFosproteins.AP-1regulatesavarietyofcellularprocesses,includingproliferation,differentiationandapoptosis,andcontributestobothbasalandstimulus-activatedgeneexpression.Itisactivatedbygrowthfactors,hormones,stress,cytokines,ROSandultravioletrADIation.Activationoccursbothtranscriptionallyandpost-translationally,andissignaledpredominantlythroughthemitogen-activatedproteinkinase(MAPK)cascade(7).ThecombinatorialdiversityofAP-1proteinsandotherinteractingfactorsappearstoinfluencehowspecificcelltypesrespondtoastimulus.
