1.WashtheGST-fusionproteinglutathione-agarosecomplexoncewith1%TritonX-100inPBS.

2.ResUSPendthecomplexin1%TritonX-100inPBS.

3.Prepareasmallcolumn(e.g.aglassorplasticSEROlogicalPipettewithtippluggedwithsiliconizedglasswool)andpourslurryintocolumn.

4.Afterthebeadshavesettled,flushthecolumnwithtwocolumnvolumesof1%TritonX-100inPBS.

5.Washthecolumnwithtwovolumesof150mMNaClin50mMTris-HCl,pH8.

6.ElutetheGSTfusionproteinfromthecolumnmaterialwith1volumeof20mMglutathioneand150mMNaClin50mMTris-HCl,pH8.

Thetotalvolumeoftheflow-throughwillbe3to5mlandshouldbecollectedin0.5mlaliquots.Approximately90%ofthefusionproteinwillbecollectedin1to2ofthesealiquotsandcanbemonitoredbygelelectrophoresisandsilverstaining.

Toremovetheglutathioneandtoconcentratetheelutedprotein,thesolutioncaneitherbesubjectedtoultrafiltration(e.g.usingaCentricon10TMunit),ortheproteincanbeprecipitatedusingtrichloroaceticacid(denaturing)orammoniumsulfate(finalconcentrationof80%,non-denaturing).
