Carbocyaninedyeslabelcytoplasmicmembranesandintracellularmembranestructuresefficientlyandpermanently(1).Theyhavebeenusedastracersincell–cellfusion(2,3),cellularadhesion(4,5),andmigration(6)applicationsduetotheirpropertiesoflowcytotoxicityandhighresistancetointercellulartransfer.However,thelipophilicnatureofthesedyesposeanobstacletouniformcellularlabeling.AlthoughstructurallyrelatedPKHdyeshavebeendevelopedandoptimizedforcelllabeling,theprocedurerequiresmultiplestepsandsubjectscellstoaniso-osmoticmannitolloADIngmedium(7,8). Avarietyofcarbocyaninedyeswithdifferentopticalpropertiesareavailable,allowingcellpopulationstobemarkedwithdistinctfluorescentcolorsforidentificationaftermixing.Doublelabelingcanidentifycellsthathavefusedorformedstableclusters.

CellBrite™NIRCytoplasmicMembraneDyesarenovelnear-infraredcarbocyaninedyesforlabelingthecytoplasmicmembranesoflivingcells.Duetotheirlongemissionwavelengths,near-infraredcellmembranestainscanbeusedtolabelcellsfornear-infraredsmallanimalimagingstudiesfornon-invasiveimagingofcellmigrationandcellhoming(9).Thedyeshavelong18-carbonhydrophobictailsandanadditionalwater-solublegroup.Theseuniquechemicalstructureelementsmakethedyeseasytodissolvewhileprovidinghighlystablecytoplasmicmembranestaining,unliketraditionalcarbocyaninedyeslikeDiI,DiO,andDiR,whichareoftendifficulttodissolveorpronetoprecipitationduringcellstaining.
