TACIandBCMAsignaltransductionpathwaythatenhancescellsurvivalAPRILandBAFF(alsocalledTALL-IandBLyS)areTNFfamilymembersthatactasligandsfortheBCMAandTACIreceptors.BothAPRILandBAFFbindtoboththeBCMAandTACIreceptorstoactivatethehumoralimmuneresponse,stimulatingBcellimmunoglobulinproductionandproliferation.BAFFisfoundasamembraneboundforminTcellsandasolubleformthatisreleasedfromthecelltostimulateBcellproliferationanddifferentiation.AsmembersoftheTNFreceptorgenefamily,BCMAandTACIinteractwithTRAFfamilymemberstotransducesignalsdownstreamtoNF-kappaBactivationandMAPkinasepathways.AbnormallyactiveBAFForAPRILsignalingmayplayaroleinautoimmunedisorderssuchaslupus.
