CathepsinD(CatD)isaubiquitouslyexpressedlysosomalaspartylproteaseinvolvedinthenormaldegradationofproteinsapoptosisandautophagy.HumanCatDissynthesizedontheendoplasmicreticulumasapre-pro-enzyme.PreprocathepsinD(412aa)iscleavedandglycosylatedtoformaninactiveprocathepsinD(392aa)andthenfurthercleavedtogenerateanactiveintermediate(348aa)single-chainmolecule.TheactiveintermediateisfurtherprocessedintomaturetwochainformofcathepsinD,thisprocessingstepiscarriedoutbycathepsinBorL.Thetwo-chainformconsistsofanaminoterminallightchain(14kDa)andacarboxyl-terminalheavychain(34kDa).Additionally,severalmoreaminoacidsareremovedfromthecarboxylterminusofthe34kDaheavychain.ProcathepsinD(pCD),secretedfromcancercells,actsasamitogenonbothcancerandstromalcellsandstimulatestheirproinvasiveandpro-metastaticproperties.Acidproteaseactiveinintracellularproteinbreakdown.InvolvedinthepathogenesisofseveraldiseasessuchasbreastcancerandpossiblyAlzheimerdisease.
