TypeIVCollagenase,72-kD,isofficiallydesignatedmatrixmetalloproteinase-2(MMP2).Itisalsoknownasgelatinase,72-kD.MMP-2playsanessentialroleinangiogenesisandarteriogenesis,twoprocessescriticaltorestorationoftissueperfusionafterischemia.MMP-2expressionisincreasedintissueischemia,buttheresponsiblemechanismsremainunknown.1Matrixmetalloproteinases(MMPs)catalyzeextracellularmatrixdegradation.Controloftheiractivityisapromisingtargetfortherapyofdiseasescharacterizedbyabnormalconnectivetissueturnover.MMPsareexpressedaslatentproenzymesthatareactivatedbyproteolyticcleavagethattriggersaconformationalchangeinthepropeptide(cysteineswitch).ThestructureofproMMP-2revealshowthepropeptideshieldsthecatalyticcleftandthatthecysteineswitchmayoperatethroughcleavageofloopsessentialforpropeptidestability.2Thegeneislocalizedto16q21usingsomaticcellhybridsandinsituhybridization.3ThestandardproductusedinthiskitisrecombinantratMMP-2,consistingof662aminoacidswiththemolecularmassof72KDa.ThedetectedMMP-2includeszymogenandactiveenzyme.
