Description:

Stainingofcellswithafluorescentlylabeledanti-gp64antibodyallowsforidentificationofinfectedinsectcells.Byinoculatingcultureswithaseriesoflogdilutionsofvirus,andstainingthecultures18–20hourspostinoculation,theratioofinfectedtouninfectedinsectcellscanbedeterminedbyflowcytometry.Statisticalanalysisofthepercentageofinfectedcellsinthevirusdilutionseriesenablesaccurateinfectioustiterdetermination.

Alternativeassaysthatalsoclaimresultsinunder24hours(suchasparticlecountinginstrumentsandqPCR)areindeedfastbutdonotdelivertrueinfectioustitervalues.Incontrasttotheplaqueassay,theculturingconditionsemployedfortheflowcytometricassaycloselyreflectactualculturingconditionsutilizedforexpressioncultures,resultinginamorerelevanttitervalueinafractionofthetime.Providedaccesstoaflowcytometerequippedtohandle96wellplates,theflowcytometricassayiseasilyadaptedtohighthroughputprocesses.
