Thesecretedhumanβisoform(teratocarcinomaderived)autotaxinwithC-terminal6-HistagwasexpressedinSf9cellsandpurifiedusingnickel-NTAchromatography.Theenzymeissuppliedlyophilized(Containsdextrose,Hepes,trehaloseandTween20)andistestedforactivitybeforerelease.

Autotaxin,alsoknownasATX,eENPP2,lysophospholipaseD,phosphodiesterase1αandplasmacellglycoprotein-1,isasecretedglycoproteinthatiswidelyexpressedwithhighlevelsintheserum.ViaitslysophospholipaseD(lysoPLD)activityautotaxinhydrolyzeslysophosphatidylcholine(LPC)togeneratethephospholipidgrowthfactorlysophosphatidicacid(LPA).Theenzyme’ssameactivityhydrolyzessphingosylphosphorylcholine(SPC)toformsphinosine1-phosphate(S1P).
