Bovine serum albumin (BSA) is a slightly acidic ~66 kd protein. BSA is commonly used in biological research due to its low cost and inertness. Dilute enzymes can be stabilized by BSA (useful for restriction digests). BSA is a common blocking agent in western blotting and ELISA s. Although, bovine serum albumin is exceptionally soluble (up to 50% w/v in buffered saline), it is typically used at a concentration of 0.1-1% w/v.

在病例条件下 肿瘤细胞在生长或转移时,病变细胞释放的化学物质和肿瘤衍生物的甲状腺激素相关肽均通过产生NO和激活KATP来降低集合淋巴管 节律泵 的活性,从而影响了淋巴的生成 转运 使得肿瘤组织淋巴回流较小,分子量相对较小的物质从肿瘤间质中清除较快,而大分子被截留,因此大分子物质还可以在肿瘤组织中选择性的聚集。高血管通透性和截留率是白蛋白作为抗肿瘤药物靶向载体的必要条件,白蛋白的这种效应符合大分子物质在肿瘤组织的透过性增强及滞留效应。

该纳米载体取材绿色、设计巧妙,可在肿瘤部位依次针对肿瘤表面过表达酶、微酸和高谷胱甘肽(GSH)浓度等微环境实现三重响应性释药(图2)。作者首先通过定向酶解奶酪副产物-乳清蛋白获得两亲性多肽,在水溶液中可自组装形成30纳米单分散胶束peptosome (PS),荷载疏水性抗癌药物姜黄素Cur,解决了姜黄素水溶液难溶的问题,并降低其对正常组织的副作用。随后他们对其表面修饰功能肽段PePm,其中Pe可靶向乳腺癌表面的ErB2受体,且兼具较好的抗癌转移效果。此外,Pm片段可以根据肿瘤表面过表达金属基质蛋白酶MMP的特异性水解,通过交联单个载体内部自身巯基形成二硫键,使PS具有癌细胞高GSH浓度响应释药的功能。他们将所构建的载体PePm/PS/Cur经尾静脉注射到小鼠体内后,经EPR效应和Pe的靶向效应在肿瘤部位高度富集,载体表面Pe肽被MMP水解脱离,使Pe肽留在肿瘤细胞表面发挥抑瘤和抗转移效果。PS/Cur继续通过 质子海绵 效应从癌细胞溶酶体中逃逸,在细胞质内通过GSH响应逐渐解体,释放出PS/Cur内荷载的姜黄素,发挥抑制肿瘤的作用。

豆球蛋白A(Con A),又名豆凝集素、豆素,是一种植物血凝素,具有强力的促有丝分裂作用,有较好的促淋巴细胞转化反应的作用,其促淋巴细胞转化zui适浓度为40-100 g/ml,能沉淀肝糖原,凝集羊、马、狗、兔、猪、大鼠、小鼠、豚鼠等动物及人红细胞。还能选择性激活抑制性T细胞(Ts)细胞,对调节机体免疫反应具有重要作用。因此,通过使用Con A来活化病态(或老年)时的Ts细胞这一途径,可望改观一些自身免疫性疾病,甚或移植物排斥反应或恶性肿瘤的防治前景。

豆蛋白(Concanavalin A,Con A)是从豆(Jack bean)中提取出来的凝集素,能沉淀多种糖类,葡聚糖和果聚糖等很多其它多糖,以及免疫球蛋白和血型物质等多种糖蛋白,还沉淀肺炎球菌多糖,并能凝集多种红细胞。Con A 能和很多细菌和动物细胞反应,能区分某些正常和肿瘤细胞,能促进细胞分裂(促有丝分裂作用)。Con A IV 用于淋巴下拨转化实验,也被用来刺激T细胞产生IL-1 样因子,还用于脾细胞增生,研究激活所必需的配体/受体作用。此外,尚区分正常分泌ACTH 的垂体细胞和分泌ACTH 的腺肿瘤细胞、用于研究糖基化在离子通道调节中的作用、用于糖蛋白的分离纯化和多糖、糖蛋白糖链结构的研究等。

Concanavalin A (con A) is a homotetrameric, 26 kDa lectin derived from the jack bean, Canavalia ensiformis. Concanavalin A binds to alpha-D glucose and alpha-D-mannose containing saccharides. Concanavalin A requires both Mn2+ and Ca2+ ions as a co-factor for binding. Since each con A has four binding sites, it can agglutinate cells such as red blood cells. Con A can also cause cell surface receptor aggregation which can be mitogenic for some lymphocytes.

链霉亲和素(streptavidin下称SA)是与亲和素(avidin下称AV)有相似生物学特性的一种蛋白质,是streptomyces avidinii菌的分泌物,其分子量及结合生物素的能力与鸡蛋清中的亲和素相似,等电点6.0,非特异性结合远比亲和素低。

链霉亲和素是四聚体蛋白,大小为66KDa。一分子链霉亲和素可以高度特异性地与四分子生物素结合,两者之间的亲和力极为强烈,链霉亲和素-生物素复合物的解离常数处于 10-14 mol/L 数量级,这一性质常用于分子生物学用途。其中一条完整的SA肽链中有159个氨基酸残基,分子量为16450

Avidin is a tetrameric protein commonly isolated from egg whites. Avidin binds to biotin with strength approaching that of a covalent bond. Disruption of the avidin-biotin bond occurs only under extraordinary conditions, such as pH 1.5 with 6 M guanidium hydrocholoride. Fluorescent avidin can bind to biotinylated antibodies used in immunohistochemistry and immunocytochemistry. Avidin thiol can bind to gold surfaces and act as a glue for biotinylated proteins. Avidin agarose is useful for efficient removal of biotinylated proteins from a liquid phase.
