Theassaydependsuponamonoclonalantibody,A717,thatspecificallyrecognizestheglycatedmoietiesinglycatedalbumin.Inpractice,standardsorsamplesareaddedtoA717coatedplates(Glycaben).Theglycoalbuminintheseanalytesbindstooriscapturedbytheantibodyonthesolidphase,andunboundmaterialsarewashedaway.Asubsequentincubationwithanenzymeconjugatedantihumanalbuminantibodyleadstodetectionoftheboundmolecules.BoundconjugateisdeterminedcolorimetricallyafterreactionwithaTMBsubstratesolution. Byrelatingsampleresponsewiththatforthestandards,theconcentrationofglycoalbuminmaybedetermined.Aseparateanalysisoftotalalbuminallowstheresultstobeconvertedtopercentoftotalalbuminthatisglycated. |
