PolyethyleneGlycolDiacrylate(PEGDA)hydrogelsarepowerfultoolsforuncoveringbasiccellularBIOLOGybecausetheyareconsideredbiologicallyinert(“blankslate”)andtheirmechanicalpropertiescanbevariedoveralargerangeofmoduli.
PEGDAisanemergingscaffoldfortissueengineeringandregenerativemedicinesincepolymerizationcanoccurrapidlyatroomtemperatureandrequireslowenergyinput,hashighwatercontent,iselastic,andcanbecustomizedtoincludeavarietyofbiologicalmolecules.
PEGDAisacrosslinkerrepeatingpolymercrosslinkerandis>80%acrylated.Itcanbeusedasathiol-reactivecrosslinkerinourHyStemsystemsatlowconcentration,orcanbeusedtomakeaPEGDA-onlyhydrogelinthepresenceofafree-rADIcalchainphotoinitiatorandlightsource.PEG-onlygelsdonottypicallysupportcellattachmentwithouttheincorporationofcellularattachmentsites.ApplicationsRegularand3-DcellcultureTissueengineeringPhotolithography

PEGDAispreparedbydissolvingthelyophilizedsolidintheDGWater.PEGDAhydrogels(1mL)shouldbepreparedinthefollowingmanner:
- AllowthePEGDAtocometoroomtemperature.
- Underasepticconditionsadd1.0mLofDGWaterto100mgofPEGDAvialfora10%(w/v)solution.
- Invertseveraltimestodissolve.Thesolutionwillbeclearandslightlyviscous.
- Tosterilize,filterthrougha0.2micronfilter.
- AdddesiredvolumeofphotoinitiatortoPEGDA.Placethevialonarockerorshakerandallowsolidstocompletelydissolve(approximately10-20minutes).
- Keepsolutionprotectedfromlightuntilreadytocrosslink.Pipettesolutionintodesiredformat(i.e.96wellplate)andphotocrosslink.
- Hydrogelpropertiesmayvarydependingontimeofexposureandtypeoflight.EnsurethePhotocrosslinkinglightisincloseproximitytothehydrogelsolution.
- FreezeunusedPEGDAsolutionat-20°Candprotectedfromlight.
