MonoclonalantibodyIIIB11recognizestheurokinase-typeplasminogenactivatorreceptor(uPARorCD87),aGPIanchoredhighlyglycosylated,single-chainpolypeptide.CD87,a45-to60-kDprotein,consistsofthreestructurallyhomologousdomainsofapproximately90aminoacids,designatedD1,D2,andD3,eachcontainingaconservedarrangementofdisulfidebondsandseparatedbyshortinterdomainlinkersequences.uPARexistsinvariousmolecularforms;heavilyglycosylated,acleavedform,asolubleformandsplicedforms.CD87isexpressedbymonocytes/macrophages,neutrophils,endothelial,smoothmusclecellsandepithelialcells.CD87isoverexpresseduponexposuretoinflammatorymediators.Theproteinisalsohighlyexpressedbytumorcellsandcontributestotheirinvasivenessandmetastaticpotential.CD87isamultiligandreceptorthatoperatesasakeyelementinpathophysiologicalprocessesinvolvingcellmigrationandtissueremodellingduringinflammationandcancermetastasis.CD87bindswithhighaffinitytheSer-proteinaseurokinase-typeplasminogenactivator(uPA)which,uponbindingcatalyzestheconversionofplasminogenintoplasminwhichactivatesvariousmatrixmetalloproteinaseleADIngtoahighpotentialforpericellularproteolysis.CD87functionsalsoasanadhesionmoleculebybindingvitronectin,thuscontributingtoleukocyticadherence.High-affinitybindingofCD87tobothuPAandvitronectinrequiresthepresenceofdomain1(D1).Yet,CD87participatesincelladherenceandmigrationalsothroughtwootherroutes,aphysicalassociationandfunctionalinteractionwithvariousintegrinsandviaanintrinsicchemotacticactivityassociatedtotheD1-D2linkersequence.MonoclonalantibodyIIIB11recognizesD2oftheurokinase-typeplasminogenactivatorreceptor(CD87).Theantibodyrecognizesaminoacidsequences125-132oftheD2domain.
