Description:MASP-3,Human,ELISAkit
Themannan-bindinglectin(MBL)pathwayofthecomplementsystemisinitiatedbythebindingofMBLorficolintospecificcarbohydratestructuresfoundonthesurfaceofavarietyofmicroorganisms.ThisbindingisfollowedbytheactivationofMBL-associatedserineproteases(MASPs).TherearefourknownMASPs:MASP-1,MASP-2,MASP-3andMap19(alsoknownassMAP,asmall,separatelysynthesizedfragmentofMASP-2).MASPsaresecretedassinglechainprecursorsandareprocessedintoaheavychain(A)andcatalyticlightchain(B)linkedbydisulfidebonds.AllMASPsformhomo-dimersandindividuallyassociatewithMBLandtheficolinsinaCa2+-dependentmanner.MASP-3isanalternativelysplicedproductfromtheMASP-1gene,containinganidenticalAchain,butanentirelydifferentBchain.MASP-3isanapproximately100kDaproteinandassociateswithbothMBLandL-ficolininthepresenceofCa2+.TheMASP3dimercanbedisruptedbyEDTA.MASP-3isexpressedmainlybutnotexclusivelyintheliver.LiverexpressedMASP-3issecretedintotheblood.UnlikeMASP-2,noproteolyticactivityoncomplementproteinssC2,C3andC4forMASP-3hasbeendetected.MASP-3exertsinhibitoryactivityonMASP-2function.However,nonaturalsubstratesforMASP-3areknown.MASP-3concentrationinhealthyhumanserumisnormallydistributedwithameanvalueof3796ng/ml.MASP-3ispresentatbirthatapproximately80%ofthelevelmeasured6,9and12monthsafterbirth.TheMASP-3levelisstablethroughouta1-yearperiod.ThehighexpressionofMASP-3inastrocytesindicatesthatMASP-3mightalsobeinvolvedinotherfunctionsthatcouldbeimportantinthebrain.Inmultiplesclerosispatients,highMASP-3levelsseemtohaveapositiveeffect(medianMASP-3levels:4539ng/ml).MASP-3deficienciesarecurrentlynotreported.
