Tumornecrosisfactoralpha(TNF-alpha),ahomotrimeric17KDprotein,isapotentmediatorofinflammatoryandmetabolicfunctions.TNF-alphawasoriginallydetectedasahighlycytotoxiccytokinefortumorcells,itcausestumornecrosisinvivoandshowscytolyticactivityagainsttumorcellsinvitro.FurThermore,TNF-alphahasbeenimpliedascentralmediatorinshockinducedbygramnegativemicro-organisms.ThecytokineTNFisfoundtobeacentralmediatorinmanyinflammatoryandimmunologicalprocesses:itcanbeinducedbyvariousproductsofmicro-organismsandbyvariouscytokinesbutitalsoinducesonitsturntheproductionofmanycytokines.SignaltransductionoccursviatwotypesofTNF-receptors,thep55(RI)andp75(RII)TNF-receptors.Thereceptorsdifferstronglyintheirintra-cellularsignalingpathways.TheTNF-alphatrimerinteractswitheitherofthetwotypesofTNF-RleADIngtoreceptorcross-linking.
