TheDetectX®AllopregnanoloneImmunoassaykitisdesignedtoquantitativelymeasureAllopregnanolonepresentinextractedserum,plasma,anddriedfecalsamples,andinurineandtissueculturemediasamples.Anallopregnanolonestandardisprovidedtogenerateastandardcurvefortheassayandallsamplesshouldbereadoffthestandardcurve.StandardsordilutedsamplesarePipettedintoaclearmicrotiterplatecoatedwithanantibodytocapturerabbitantibodies.Anallopregnanolone-peroxidaseconjugateisaddedtothewells.ThebindingreactionfortheEIA/ELISAisinitiatedbytheadditionofapolyclonalantibodytoallopregnanolonetoeachwell.Afterincubationtheplateiswashedandsubstrateisadded.Thesubstratereactswiththeboundallopregnanolone-peroxidaseconjugate.Afterashortincubation,thereactionisstoppedandtheintensityofthegeneratedcolorfromtheEIA/ELISAisdetectedinamicrotiterplatereadercapableofmeasuring450nmwavelength.
