BackgroundVoltage-dependentcalciumchannels(VDCCs)arelarge(>400kDa)heteromerswhichcontain,minimally,threecoresubunits1,2/,ina1:1:1stoichiometry1.ExpressionofVDCCgeneproductsinXenopusoocytes,ortransfectedcellsshowsthatthe1subunitscontaintheionchannelporewhiletheauxiliary2/andsubunitsconferoptimalcellsurfaceexpressionandchannelkinetics1.Untilrecently,theonlyexceptiontotheaboveparADIgmwastheskeletalmuscleVDCC,which,inadditiontothe1,2/,coremotif,alsohasanadditionaltightlyassociatedintegralmembraneglycoproteinsubunittermed1.Uponco-expressionwiththe1.1,2/1,1asubunitsoftheskeletalmuscleVDCC,subunitsalterthepeakcurrents,andthekineticsofchannelactivationandinactivationwiththeoveralleffectbeinganormalisationofcurrentstothoseresemblingtheendogenouschannel2.Together,theseresultssuggestthatsubunitsmodulateskeletalmuscleVDCCsbystABIlisingtheirconformation.The2subunits,orstargazin,theproductofagenemutatedinthestargazermouse,isahomologofthe1channelandisselectivelyexpressedinthebrain.ItisconsideredtobeaputativeneuronalCa2+channelsubtypemainlybasedonitshomologytothe1.
