CD28andCTLA-4togetherwiththeirligands,CD80(B7-1)andCD86(B7-2),constituteoneofthedominantcostimulatorypathwaysthatregulateTandBcellresponses.CD28andCTLA-4arestructurallyhomologousmoleculesthataremembersoftheimmunoglobulin(Ig)genesuperfamily.BothCD28andCTLA-4arecomposedofasingleIgV-likeextracellulardomain,atransmembranedomainandanintracellulardomain.CD28andCTLA-4arebothexpressedonthecellsurfaceasdisulfide-linkedhomodimersorasmonomers.Thegenesencodingthesetwomoleculesarecloselylinkedonhumanchromosome2andmousechromosome1.MouseCD28isexpressedconstitutivelyonvirtually100%ofmouseTcellsandondevelopingthymocytes.CellsurfaceexpressionofmouseCD28isdownregulateduponligationofCD28inthepresenceofPMAorPHA.Incontrast,CTLA-4isnotexpressedconstitutivelybutisupregulatedrapidlyfollowingTcellactivationandCD28ligation.CellsurfaceexpressionofmouseCTLA-4peaksapprox.y48hoursafteractivation.AlthoughbothCTLA-4andCD28canbindtothesameligands,CTLA-4bindstoB7-1andB7-2witha20-100foldhigheraffinitythanCD28.CD28/B7interactionhasbeenshowntopreventapoptosisofactivatedTcellsviatheupregulationofbcl-XL.CD28ligationhasalsobeenshowntoregulateTh1/Th2differentiation.
