Transthyretin(TTR,55kDahomotetramer)isathyroidhormone-bindingproteinthattransportsthyroxine(T4)fromthebloodstreamtothebrain.TTRwasoriginallycalledprealbuminbecauseitranfasterthanalbuminonelectrophoresisgels.TTRisproducedintheliverandcirculatesinthebloodstream,whereitbindsretinolandthyroxine.TTRissuggestedthatplaysanessentialroleinbrainfunction.Theproteinconsistsofaround130aminoacids,whichassembleasahomotetramerthatcontainsaninternalchannelinwhichT4isbound.Withinthiscomplex,T4appearstobetransportedacrosstheblood-brainbarrier,where,inthechoroidplexus,thehormonestimulatesfurthersynthesisoftransthyretin.Theproteinthendiffusesbackintothebloodstream,whereitbindsT4fortransportbacktothebrain.TTRisknowntobeassociatedwiththeamyloiddiseasessenilesystemicamyloidosis(SSA),familialamyloidpolyneuropathy(FAP),andfamilialamyloidcardiomyopathy(FAC).Thyroidhormone-bindingprotein.Probablytransportsthyroxinefromthebloodstreamtothebrain.
