Peroxiredoxin(Prx)isagrowingperoxidasefamily,whosemammalianmembershavebeenknowntoconnectwithcellproliferation,differentiation,andapoptosis.Manyisoforms(about50proteins),collectedinaccordancetotheaminoacidsequencehomology,particularlyamino-terminalregioncontainingactivesitecysteineresidue,andthethiol-specificantioxidantactivity,distributethroughoutallthekingdoms.Amongthem,mammalianPrxconsistsof6differentmembersgroupedintotypical2-Cys,atypical2-CysPrx,and1-CysPrx.ExceptPrx6belongingto1-CysPrxsubgroup,theotherfive2-CysPrxisotypeshavethethioredoxin-dependentperoxidase(TPx)activityutilizingthioredoxin,thioredoxinreductase,andNADPHasareducingsystem.MammalianPrxsare20-30kilodaltoninmolecularsizeandvaryinsubcellularlocalization:Prx1,2,and4incytosol,Prx3inmitochondria,Prx4inERandsecretion,Prx5showingcomplicateddistributionincludingperoxisome,mitochondriaandcytosol(1).
