Calciumbindingproteins(CBPs)regulateintracellularlevelsofcalciumCa2+ions.CBPsareinvolvedinnumerousfunctions,suchascellsignaling,calciumuptakeandtransport,andcellmotility.Calretinin(CR),calbindinD-28k(CB)andparvalbumin(PV)belongtothelargefamilyofEfhandcalcium-bindingproteins.TheEF-handisanaminoacidsequencewithaharacteristicthreedimensionalhelix-loop-helixstructurewithhighaffinityforCa2+.CalbindinD28k(calbindin)isahexaEF-handproteinof261residueswith59%sequenceidentitytocalretinin.Itisasingle-domainproteinandthesixEFhandsinteractextensivelywithoneanothersothattheyformonelargeglobulardomain.Calbindinisfoundinthebrainandcentralnervoussystemwithadistinctdistributionovercertainneuronalsubtypes.Neuroprotectiveeffectsofcalbindinhasbeenreportedinischemicandglutamatetoxicitymodels,primarilyduetoitsabilitytochelatecalcium.MicelackingcalbindinhavebeenshowntosufferfromimpairedmotorcoordinationduetoalteredneuronalCa2+-homeostasis.Notably,calbindinisfoundinhighconcentrationinthekidneyandinpancreaticβ-cells,whereithasbeenshowntoprotectagainstapoptosis.Inosteoblasts,anti-apopticactivityofcalbindinthroughthebindingtocaspase-3hasalsobeenreported.Calbindinprotectsagainstapoptoticandnecroticcelldeath,suggestingitsabilitytobuffercalcium.CalbindinhasbeenreportedtobindsZn2+,revealingnegativeallostericeffectbetweentheZn2+-andCa2+-bindingevents.Calretininisacalcium-bindingproteinwhichisabundantinauditoryneurons.
