Theubiquitinproteasomepathway(UPP)isthecell’sprinciplemechanismforproteincatabolism.TheUPPhasbeenshowntohavesignificantinvolvementintheregulationofcriticalcellularprocessessuchastranscription,oncogenesis,cellcycleprogression,development,growth,selectiveeliminationofabnormalproteinsandantigenprocessing.Theproteasomeisalarge,multimericproteasethatcatalyzesthefinalstepoftheUPPintracellularproteindegradation.Theproteasomeexistsinmultipleformswithintheeukaryoticcell,andcontainedinallisoformsisthecatalyticcoreknownasthe20SProteasome.The20Sproteasome(700kDa)isarrangedasfouraxiallystackedheptamericringswithtwoβ-subunitringssandwichedbetweentwoα-subunitrings.Themulticatalyticcentersarelocatedwithintheinternalcavityoftheβ-subunits.The26SProteasomeisdefinedbyhavingoneortwo19Sregulatorycapsonthe20Scoreparticle.Itisthe26SthatisresponsIBLefortherecognitionandsubsequentdegradationofpoly-ubiquitinatedproteinsintosmallerpolypeptides.ThisdegradationisubiquitinandATPdependentandwillnotbe26SProteasomemediatedunlessthetargetsubstrateisfirstubiquitinatedviatheconjugationcascade.
