BACKGROUND

FactorVIII(FVIII)isanessentialblood-clottingprotein,alsoknownasanti-hemophilicfactor(AHF).Inhumans,factorVIIIisencodedbytheF8gene.DefectsinthisgeneresultinhemophiliaA,arecessiveX-linkedcoagulationdisorder.FactorVIIIisproducedinliversinusoidalcellsandendothelialcellsoutsideoftheliverthroughoutthebody.Thisproteincirculatesinthebloodstreaminaninactiveform,boundtoanothermoleculecalledvonWillebrandfactor,untilaninjurythatdamagesbloodvesselsoccurs.Inresponsetoinjury,factorVIIIisactivatedandseparatesfromvonWillebrandfactor.Theactiveprotein(sometimeswrittenasfactorVIIIa)interactswithanothercoagulationfactorcalledfactorIX.Thisinteractionsetsoffachainofadditionalchemicalreactionsthatformabloodclot.
