The26Sproteasomeisthecentralproteaseinubiquitin-mediatedproteindegradation.Itisalargemulti-proteincomplex,whichplaysafundamentalroleincellularhomeostasisbycontrollingdegradationofkeyproteinsthatareinvolvedinapoptosis,cellcycle,andsignaltransduction.The26SproteasomeisresponsIBLeforcellqualitycontrolbyeliminatingmisfoldedproteinsfromthecytosolandendoplasmicreticulum.Withitsdiversefunctions,theproteasomeisanimportanttargetfordrugdevelopment,particularlyincancerandneurodegeneration.

Anumberofselectiveproteasomeinhibitorshavebeenidentifiedinthelasttwodecadesandapprovedforclinicaluse(bortezomib,carfilzomib).Severalotherproteasomalinhibitorsarecurrentlyintrialsfortreatmentofmultiplemyelomaandothermalignancies.The26Sproteasomeiscomposedofasinglecatalytic20Scomplex(coreparticle,CP)andoneortwo19Sregulatorycomplexes(regulatoryparticles,RP).The19Sregulatorycomplexisresponsiblefordeubiquitylation,unfoldingandre-directionofproteinstothecatalyticsiteswithinthe20Scomplex.
