Background:Thegreenfluorescentprotein(GFP)iscomposedof238aminoacidresidues(26.9kDa)andexhibitsbrightgreenfluorescencewhenexposedtoultravioletbluelight.IthasbecomeawellestablishedreporterforgeneexpressionandMarkerforproteintargeting.SubtlemodificationshavebeenmadetothesequenceofGFPimprovingitssolubilityandphotostablility(eGFP)toallowacquisitionofmorecomplexdata(FRET,FRAP).Moreover,subtleresiduemodificationshavebeenengineeredtoaltertheemissionwavelengthsuchasinCFP,YFPtofacilitateduellabelling.Anti-GFPAbisapolyclonalantibodyraisedinrabbitbyimmunisationwith,andaffinitypurificationagainstfulllengthGFP(Faixetal.,2001).ItproducesexceptionalstainingforGFP,PA-GFP,EGFP,CFPandYFPinwesternblottingatadilutionof1:10,000andbioimagingapplicationsathighdilutions,ithasfeaturedinanimpressivenumberofpublications.
