Overview

TheMouseTumorNecrosisFactor-alpha(TNF-α)ELISAKitisdesignedforthequantitativedetectionandmeasurementofmouseTNF-αinBIOLOGicalfluidssuchasserum,plasma,andcellculturesupernatants.TNF-αisprimarilyproducedbyactivatedTcellsandmacrophagesinresponsetoinflammationorinfection,butisalsoproducedbyBcells,NKcells,mastcells,neutrophils,dendriticcells,microglia,endothelialcells,smoothmusclecells,cardiomyocytes,andfibroblasts.TNF-αisapro-inflammatorycytokineandstimulatesanti-tumorresponsesinvitro.TheassayisbasedonthesandwichELISAmethod,inwhichsamplesareaddedtoELISAstripplatespre-coatedwithcaptureantibodiesspecificforthecytokine.Thecapturedcytokineisdetectedbyadditionofabiotinylateddetectionantibody,followedbystreptavidin-horserADIshperoxidase,whichbindsthebiotinylatedantibody.Additionofthechromogenicenzymesubstrate3,3’,5,5’tetramethylbenzidine(TMB)resultsinacoloredproductwithanintensitydirectlyproportionaltotheconcentrationofcytokineinthesample.Theconcentrationofthecytokineisdeterminedbycomparisontoaserialdilutionofthecytokinestandardanalyzedinparallel.
